Firefly was really the first one that hit me hard, because I was watching it when it was first broadcast.
Since then I'd have to say:

There are several others I had to mourn after the fact, not seeing some of them until they repeated on the Sci-Fi Channel or I caught them on DVD:

Freaks and Geeks
A show very dear to my heart was broadcast back in the Dark Ages of 1970. There were no VCRs then, much less DVDS, Netflix or Hulu. It has yet to be released on any video format so it has been nearly 40 years since I've seen it, but if "My World and Welcome To It" is ever released I'll be placing the first order.
There really are way too many series that fit this category. Curse you, Chris, for bringing back all the pain.

Yeah, as soon as NBC canceled it, TNT picked it up. They re-aired the first season back in January. Then season 2 in the spring/summer. Season 3 should start early next year.

Yeah, I knew that, and caught all the remaining eps on TNT. I just hadn't heard that they'd actually ordered MORE. That's pretty freaking awesome.
Cast still intact, I hope? Kevin Alejandro is a friend of the family - he went to school with my niece in Snyder, Texas, so I like to see him do well.

Good call on Carnivale ECGordon.
I wasn't watching Firefly when it was first on, unfortunately, but that definitely hurt the most.
Second was definitely Carnivale though. Again, I watched it after it was on TV, but didn't realise it was cancelled until I was about halfway through season 2. Such a fantastic show.

Firefly. Though at the time, it was more of a hey... baseball? where is my show??? insert me checking the internet - not canceled moved - but may be on teh chopping block, followed by campaigning to save the show etc. About 6 months after the show was canceled, it really hit me that it was gone. I never thought I would describe something I saw on TV as a crushing blow, like losing a friend, but that is how it felt. It did lead me to an incredibly shiny group of people, and some wonderful and amazing experiences. Browncoats are a second family!
Pushing Daisies is a distant second. I was excited at the directions this show was going in, and despite its morbid theme it was an incredibly cheery fairy tale I looked forward to every week. It brought this shiny optimism that left me smiling.
I don't know that any show will ever strike such a chord with me the way Firefly did. Mal was right, "Once you've been to Serenity Valley, you never leave."

Ok, yes, it wasn't all that good - but I liked it better than Deep Space 9 and Babylon 5, the two shows it was up against, and got creamed by, cause it was more up close, personal, grittier...
Some of that flavor, by accident or design, leaked into Firefly, which was one of the things that drew me to it to begin with.
Not that DS9 and Bab5 were all that bad, in fact I kinda liked Bab5, although I have "issues" with the world of 'trek' - but it was too distant, too impersonal, and I couldn't really connect with any of the characters save Lando Molari, who I didn't like that much, but understood all too well.
The other one was Space: Above and Beyond.

That was some good stuff, and I remember often pushing a couple limits on the way home, cause with my work schedule I would usually miss the first ten-fifteen minutes of it anyways...
And then they wiped it, I was pissed, cause I was already prettymuch in the middle of dumping TV from my life thanks to the OJ Circus, so that was one more nail in the coffin.
