Of the ones already posted I'd agree with:
Star Trek II: The Wrath Of Khan - Is there a greater scifi death scene/funeral?
Pan's Labyrinth - So sad but so hopeful depending on your pov.
Up - It shows the greatness of Pixar films that they can get to you so early in a film.
Powder - Fantastic, very underrated film. Very moving.
I'll add:
E.T. - On my first viewing.
Switched At Birth - One of those true story movies. Got me when the Dad had to tell his daughter that she wasn't his daughter.
The Green Mile - Gets me every single time.

Oh, snap - I forgot "To Sir, With Love". I *LOVE* that movie!
You should really check out "Goodbye, Mr. Chips", too. Either the 1939 Robert Donat version or the later Peter O'Toole version - they're both fantastic. I prefer the later one myself, though.
And yes on Gump.
And Chris, I didn't hate Steel Magnolias. It's a chick-flick, sure, but I didn't hate it, and I expected to.
