Well she kicked the caretakers ass for one, and play the games, half the kills the Master Chief gets, she tells him, so tecnicaly the kills go to her to, and that count is in the thousands, so..........

Well, River, of course (she did it without guns, remember, 99% of the time!), and personally I'd say Ripley #2.
Buffy? She was a woos...yeah, so she killed vampires, but she had SUPER STRENGTH to do it and still got her ass kicked a lot. Besides, she spent seven years doing nothing but moaning about not having a life; that's a strong woman? Pffft.
Faith comes closer, but I don't like her politics or attitude.
River is a "pure" heroine...how many of those are running around? You ARE speaking of best "ass kicker", not strategist, thinker, plotter, etc., so it has to be River.
