To me, it's a three-way tie for 3rd likely to die. Jayne could have evolved as a character enough to give up his life for his crew by the time a second film is set. And Simon would definitely lay down his life for River or Kaylee if they were in danger.
Mal
One of the major draws of the BDS and BDMs (if we get more than one). Likely to be the last of the original BDMs to die and doing something big to save the lives of those he cares about.
River
One of the other major draws of the BDS and BDMs. Her death scene would be on par with the The Gift for the sheer scale and brilliance of how her death is handled. Probably would die at the end of the first BDSequel to provoke the events of the second BDSequel that would complete the Bedlam Bards' desired Big Damn Trilogy
