I'm the third person, I believe, to post something similar to this, so I won't quote it again, but my favorite scene from Serenity is the opening scene (after River's escape and The Operative, of course) between Mal and Wash. My best friend and I have learned the whole scene and we quote it at random in the middle of our conversations. I love it!

My favorite has to be one of Kaylee's from TBDM when Zoe, Jane, Simon, River and Kaylee are about to make a stand against the reavers.
Simon (to Kaylee): I just wanted to keep River safe. Spent so much time on Serenity ignoring anything ... that I wanted for myself. My one regret in all of this ... is never being with you.
Kaylee: With me? You mean to say, as ... Sex?
Simon: I mean to say.

In Bushwhacked, during the montage of interrogation scenes, when it cuts to Jayne, and he is just sitting with his arms crossed glaring at the Alliance guy. I crack up every time.

I love that whole part. Each person's interrogation fits their character so well.
Jayne's is the best, though. I always crack up, too.
