My favourite character? Either Mal or Jayne (I really can't choose between them).
They're both just loveable rogues really.
Book: “A duel?”
Wash: “With swords?”
Simon: “The Captain's a good fighter, he must know how to handle a sword?”
Zoe: “I think he knows which end to hold.”

I love Shepherd Book.
Ron Glass puts it best, "He's somebody who's looking after humanity, caring for people. But he has another side -- he'll kick your ass, too."

Inara: Who's winning?
Simon: I can't really tell, they don't seem to be playing by any civilized rules that I know.

For me, it would have to be Kaylee. She is just so alive ... and is the emotion of the ship.
-- Cap'n, can you stop Kaylee from being so cheerful.
Ain't no power in the 'verse that can do that.

Well....I would definitely say that in the realm of sci-fi shows the entire crew of Serenity would be my favorite characters. The interaction between them reminds so much of the interaction between my & my friends that it's eerie. I guess that's part of why I like the show so much.
As for favorite character...I guess I would have to say Captain Reynolds. I can relate to his character and apparently have a tendency to act like him. A fact that my friends never miss a chance to call me on when I do it.

River.
Summer Glau could have been a huge silent film star. She makes poetry with her body. Just watching her walk through Serenity in OIS is like watching a dance recital. Or check out the way she looks when she's staring at the stars in Bushwhacked. Or the fascinated delight in her eyes and the way her mouth is mumbling calculations as she tries to figure out the rules ot the game the crew is playing in the same ep.
See her do the same thing in Safe when she's figuring out the dance steps and then watch the utter joy she takes in dancing and, while she's at it, improvising an even better dance than they're doing, before skidding to a confused, horrified stop as she intuits what's happening to Mal and Book. All of the above examples are wordless, and yet she's expressed more character in those scenes than actresses on other shows have with reams of dialog.
Most of the roles on Firefly are stereotypes who are in the process of being fleshed out. River is the Crazy Waif. But Summer and the writers have given River so many shades that she's way beyond that already. Naive, scary, brilliant River. Sweet and bratty little sister, confused and lost young woman, eerie and dangerous wild card. Poignant. Hilarious.
That's quite a range for someone whose dialog is so sparse and often incoherent. So. hats off to you, River. I can't wait to se what you bring to the movie. I know it'll be great.

After reading Shambleau's highly articulate post I find that I must agree with her in several respects.
Even though she is not my "favoritest" character River/Summer Glau is a very interesting and complex character being played by a very beautiful and skilled actress/dancer. It's hard to believe that this is only Summer's second(?) acting job. She is able to communicate so much through her movements and expressions that even some veteran actors would be hard pressed to compete. As Shambleau mentioned in the EP "Safe" River goes from moody & melancoly to lit up with joy in such a way that you can't help but feel joy right along with her. Her smile alone was enough to lighten my heart.
That being said....With regards to the descriptions of Rivers movements I find I must agree with Kelso who said..."I'll be in my bunk."

I think Mal's cool, because he's got so much willpower. Like in Out of Gas or when he gets taken by Neska. He always tries to do the right thing, and never gives up.

I agree with Werzbowski. Also, I think River is probably the toughest character to play. In that the script requires a lot of Glau's own interpretation and finding the right mind set for a girl who's mind is unstable. Summer Glau is doing a great job with the character! However, my fave is still Mal hands down!
