Ok, let's see. Please pardon any paraphrasing or slight misquotes. These are as they come off the top of my head not in any ranked ordrer.
Zoe's description of whate Reaver's will do to you.
Wash's "Who's flying this thing? .. That would be me.
Book telling Mal about the "Special Hell." i fee lthe scence is really made when he sticks his head around the corner and restates... "A special Hell."
Mal to Saffron: "If some one tries to kill you, you try to kill them right back."
Jubal Early: "What? Am I a lion. No, but I have a mighty roar."
Jubal Early to Simon: "You should be shot. Just to know what it feels like. They make physcologists be analyzed.."
Wash describing Zoe: "Have you ever had a 'Warrior Woman.' It's the legs, no the part just above the legs, and the part above that ..."
Wash and Zoe in OIS: W: "That sounds like Science Fiction."
Z: "We live on a spaceship dear."
Mal: ..."Or by my pretty floral bonnet, I'll end you."
Jayne: "The living legend needs some eggs."
Well that's the first ten I could think of.

I can't put them in an order, but, here are my favorites:
1. Defining moment for Shepherd Book: in the aired pilot, when Book is being cleaned up by Inara after his beating by Dobson, Book says, "I've been out of the Abbey two days, I've fallen in with brigands, beaten a lawman senseless, watched the Captain shoot a man I swore to protect. I don't even know if I think he was wrong. I think I'm on the wrong ship." Inara says something like, "Or maybe you're right where you need to be," as she places her hand on his bowed head as if she's giving him benediction. Very poignant and perfectly fits why he stays on board.
2. Defining moment for Wash: he displayed his mettle in War Stories. Zoe just bought him away from Niska, leaving Mal. Wash is telling her he's crazy. Zoe seems to think he means Niska, but Wash continues and talks about how Mal wouldn't break and kept him from breaking. Wash says he wouldn't have made it, then realizes that Niska will kill Mal. Zoe tells him that Niska will draw it out, for days if he can. Wash gets up with purpose and says, "Bastard's not gonna get days." Gives me chills every time.
3. Defining moment for Zoe: the oft-cited line about Reavers in Serenity. "If they take the ship, they'll rape us to death, eat our flesh, and sew our skins into their clothing. And if we're very, very lucky, they'll do it in that order." She delivered that line so well it still scares me, every time. Couldn't bring myself to use it for a signature.
4. Defining moment for River: her exchange with Mal over the intercom in Objects in Space. "I know you have questions." Mal, "That would be why I just asked them." River, "There's no time, Captain, I need you to trust me." Mal, "Am I dreamin'?" River, "We all are." Mal rolls his eyes. River, "Don't make faces." Very serious and very with it.
5. The exchange in The Train Job that got me in front of the TV to watch Firefly. Mal, "Whatever happens, remember that I love you." Zoe, "Sir?" Mal, "Because you're my wife." Zoe, "Right, Sir. Honey."
6. Defining moment for Mal: Ariel. After getting Jayne in the airlock. "You turn on any of my crew, you turn on me."
7. Defining moment for Simon Tam: realizing he can't fight the whole town who wants to burn River near the end of Safe, he wraps his arms around her, looks out at the crowd and says, "Light it."
8. Defining moment for Kaylee Frye: Serenity. "Machines got workin's and they talk to me."
9. Defining moment for Inara: The Train Job. Book says he feels awfully uselss. Inara tells him he could pray they make it back safely. Book says he doesn't think the Captain'd like that very much. Inara says, "Don't tell him. I never do."
10. Defining moment for Jayne: Ariel. When Mal has him in the airlock and has got it out of him that he did turn on the Tam's, this saves him: "Don't tell 'em what I did."
(This is really hard, folks. I'd quote the whole episode of Out of Gas if I could. All about Mal and Serenity. "Freedom is what...Never have to be under the heel of nobody ever again. No matter how long the arm of the Alliance gets, we'll just get a little further." "I wasn't askin', I was tellin'." "Time we stopped focusing on what we ain't got and think about what we do." "You all gonna be here when I wake up?")

I liked a Zoe one from Bushwhacked, where Jane goes into some details about the reavers, and Zoe interrupts him: "Jane, you´re scaring the women!"
Also a funny one, from "The Train Job":
Jane: "You know what the chain of command is? - That´s the chain I´ll go get and beat you with until you know who´s in rotten command here!"

I am here now Teela... I had quite the night and awoke with a strange heaviness in my head. So after much napping and water, I'm back in the saddle again. I didn't make that one up for sure... What's this about mice getting to my thought first?

Note: Hard to explain without reading.
In the Hitchikker's Guide, the smartest beings on earth are not humans, but mice. In the end, you meet up with the last two mice in existence, since earth was destroyed. Anyway, they were about to take out Arthur's brain to find the Question to the Answer, but Arthur and the rest escape. So the mice make up the question "How many roads must a man walk down?" And that is what I mean.
