Just saw something on Machinima that says they support Fillion for the role.

Uncharted 1, great game. Uncharted 2, possibly the best game I've ever played in my near two decades of game playing. And a huge part of that is because of the main character's charisma and humor, two elements that our captain will bring to the big screen. If he somehow gets cast as Nathan Drake, I will be so elated to see the union of my Firefly love with my Uncharted love.
And I hope Emily Rose, star of Haven and original voice actor for the lead female role, gets asked to come back in the movie as well.
