I would have to say that after River tells him she can "kill him with her brain" which I believe was in Trash (? correct me if I'm wrong please) Jayne might finally have caught a clue lol

Personally, I kinda like the idea that Jayne figured out River's secret, but nobody paid any attention to him just 'cause. . . well, he's Jayne.
